Tele's have worked fine that way for 50+ years. Go with a "thicker" set of pup's if you are worried about depth to your tone. EMG's if you don't mind actives, Seymore Duncans or DiMarzios if you only want to use passives.

Can't you just drill 6 holes in the base plate so it can be used either way ?

Here's my latest idea (for making a full-sized strat into a "travel guitar"). Holes in the baseplate and through the body are big enough for the strings to go through from the top of the guitar and out the back, then you run a metal rod through the ball-ends of the strings to anchor them against the back of the guitar. Then, you can always lossen the strings, pull the metal rod out and get the strings off the body. Either that or little washers with a notch cut through , so they slip onto the ball ends.
